Laccase of Streptomyces cyaneus with 6xHis Tag and 6xLys Tag
Streptomyces cyaneus strain CECT 3335 laccase (sclA) gene. Coding region for a laccase of S cyaneus strain. These enzymes are known for its ability to degrade lignin. They often got a broader substrate spectrum which allows degradation of other phenolic compounds. Hence laccases are capable of degrading antibiotics, drugs and other substances which tend to remain undegraded by common wastewater treatment techniques. This laccase showed good results regarding activity (in ABTS-Assays) and stability and showed ability to degrade Diclofenac. This laccase could be a good addition to enzymatic wastewater treatment since there are often applicated fungal laccases which are often more expensive regarding costs and time of expression. This part includes a 6x his tag c-terminal as well as a 6x lys tag n-terminal to the protein coding region. The his tag should allow purification while the lys tag could possibly improve the immobilization process in Mesocellular silica Foam (MCF) through providing additional amino-groups.
